Enclomiphene is the trans-isomer of clomiphene citrate, isolated from the mixture. Clomiphene as normally supplied is roughly 62% enclomiphene and 38% zuclomiphene — two isomers with meaningfully different behaviour, which is the entire reason for separating them.

Zuclomiphene is the more estrogenic isomer and has a half-life measured in weeks, so it accumulates with repeated administration. Enclomiphene is the anti-estrogenic isomer and clears far faster. Isolating it was intended to keep the pituitary-stimulating activity while removing the component responsible for estrogenic accumulation.

Research background

Selective estrogen receptor modulation

Enclomiphene acts as an antagonist at estrogen receptors in the hypothalamus and pituitary. Because circulating estradiol normally exerts negative feedback on gonadotropin release, blocking that signal removes the brake — the pituitary reads the situation as low estrogen and increases output.

LH and FSH response

The measured consequence is increased luteinising hormone and follicle-stimulating hormone, and downstream of LH, increased endogenous testosterone production in the testes. The compound does not supply testosterone; it stimulates the axis to make more.

Contrast with exogenous testosterone

This upstream mechanism is the reason enclomiphene appears in the fertility literature. Exogenous testosterone suppresses LH and FSH through the same feedback loop and with it spermatogenesis. Studies have examined whether stimulating the axis raises testosterone while preserving sperm parameters, which testosterone replacement does not.

Isomer separation rationale

A distinct strand compares enclomiphene against unfractionated clomiphene, testing whether removing zuclomiphene changes the profile as intended — this is a pharmacological question about the separation itself rather than about the target.

Storage and stability

Oral dosage forms are considerably more forgiving than lyophilized peptides. Capsules and tablets are stable at room temperature and require no cold chain, in transit or in storage.

Moisture is the real enemy rather than temperature. Keep the bottle tightly closed, leave the desiccant sachet in place, and avoid storing in a bathroom or anywhere humidity swings. Protect from direct light.

Purity and verification

Every lot is tested by an independent laboratory before release. HPLC establishes purity as a percentage of total content, separating the target compound from synthesis by-products; mass spectrometry confirms molecular identity.

For an encapsulated product there is a second question a vial does not raise: content uniformity. A capsule can be the right compound at the right purity and still be filled inconsistently, so Enclomiphene is checked for fill weight consistency across the batch as well as for identity and purity.
