How the maths works

Concentration = peptide mass ÷ diluent volume. A 10mg vial reconstituted with 2mL of bacteriostatic water gives 10 ÷ 2 = 5 mg/mL. Adding more water gives a lower concentration in the same total amount of peptide — the quantity of peptide in the vial never changes, only how dilute it is.

Lyophilized powder adds negligible volume, so the diluent volume and the final solution volume are treated as the same. That approximation holds at these scales.

Handling notes

Add diluent slowly down the inner wall of the vial rather than directly onto the powder, and swirl gently rather than shaking — mechanical shear and aeration both degrade peptides. A correctly reconstituted solution is clear and colourless.
